چکیده

The recently published study of peripheral blood lymphocyte subpopulations in patients with acute immunodeficiency syndrome, by Murray et al,’ may have contributed additional insight into normal T cell ontogenesis. However, the data on terminal transferase (TdT) must be interpreted with caution. It is stated that TdT levels are especially elevated in acute lymphocytic leukemia (ALL) of T cell origin. The concentration of TdT is actually lower in T cell than in null cell ALL.2’3 Furthermore, the enzyme is not detectable in mitogen-activated normal lymphocytes by biochemical assay4 or by Western blotting5; the report of putative TdT expression in such cells, as determined by indirect immunofluorescence (IF),6 may be a false-positive identification of TdT antigen.’ Murray et al purport to measure ‘cytoplasmic TdT’ with a kit supplied by Bethesda Research Laboratories (BRL). It should be noted that, at least in fresh human lymphocytes (whether normal8 or malignant9), TdT is predominantly a nuclear enzyme, as measured by immunoassay. Moreover, problems attendant on the use of the BRL kit have been described.’0

Track detection on the cells exposed to high LET heavy-ions by CR-39 plastic and terminal deoxynucleotidyl transferase (TdT)

Background: The fatal effect of ionizing radiation on cells depends on Linear Energy Transfer (LET) level. The distribution of ionizing radiation is sparse and homogeneous for low LET radiations such as X or γ, but it is dense and concentrated for high LET radiation such as heavy-ions radiation. Material and Methods: Chinese hamster ovary cells (CHO-K1) were exposed to 4 Gy Fe-ion 2000 keV/...

A new solid-phase immunoassay for terminal deoxynucleotidyl transferase: analysis of TdT antigen in cells, plasma, and serum.

A solid-phase immunoassay for terminal deoxynucleotidyl transferase has been developed using a primary antibody-coated polystyrene bead and secondary antibody conjugated with horseradish peroxidase. The immunoassay was compared with assays for enzyme activity and detection of antigen with immunofluorescence using cells from peripheral blood and bone marrow from patients with leukemia or lymphom...

Terminal deoxynucleotidyl transferase: the story of a misguided DNA polymerase.

Nearly every DNA polymerase characterized to date exclusively catalyzes the incorporation of mononucleotides into a growing primer using a DNA or RNA template as a guide to direct each incorporation event. There is, however, one unique DNA polymerase designated terminal deoxynucleotidyl transferase that performs DNA synthesis using only single-stranded DNA as the nucleic acid substrate. In this...

Sensitive and homogeneous microRNA detection using branched cascade enzymatic amplification.

This assay, termed branched cascade enzymatic amplification (BCEA), can be a novel and straightforward method for sensitive and specific microRNA detection in crude cellular extracts of cancer cells at physiological temperature, by coupling two ordinary polymerases, Klenow fragment exo(-) and terminal deoxynucleotidyl transferase.

Occurrence of a terminal deoxynucleotidyl transferase-like activity in N-2-fluorenylacetamide-treated rat liver.

A new enzyme activity that resembles terminal deoxynucleotidyl transferase (TdT-like activity) was found in rat liver. Upon treatment of rats with a hepatocarcinogen, N-2-fluorenylacetamide, the TdT-like activity increased in parallel with DNA polymerase Ci and 'Y. By phosphocellulose column chromatography, the TdT-like activity was separated from DNA polymerase Ci, fJ, and 'Y. With a poly(dA)-...

Demonstration of terminal deoxynucleotidyl transferase in thymocytes by immunofluorescence.

The cellular and subcellular distribution of terminal deoxynucleotidyl transferase (DNA nucleotidylexotransferase; nucleosidetriphosphate:DNA deoxynucleotidylexotransferase, EC 2.7.7.31) in thymocytes and peripheral lymphocytes from rat, mouse, and calf was studied by immunofluorescence using rabbit antiserum to homogeneous transferase from calf. Terminal transferase was readily detected in app...
